1. Growth Hormone Synthesis
GHRP-2's main area of research is its potential to induce GH secretion.
Mechanism: The peptide is believed to activate GHS-Rs on the pituitary gland, which may trigger a cascade of intracellular events involving G-proteins, phospholipase C (PLC), and the release of calcium ions, ultimately leading to the secretion of stored GH. This process may also involve the activation of cyclic AMP (cAMP) to further enhance GH synthesis.
Studies: Clinical trials have suggested that GHRP-2 may induce a significant increase in peak GH levels, with one trial reporting a 181-fold higher spike in GH production. Prolonged exposure was also suggested to increase GH levels by 3- to 5-fold and raise insulin-like growth factor-1 (IGF-1) levels.

GHRP-2 is posited to influence appetite by activating ghrelin receptors in the nervous system.
Mechanism: By engaging GHS-Rs, GHRP-2 may enhance the synthesis of hunger-promoting neuropeptides like Neuropeptide Y (NPY) and Agouti-related peptide (AgRP) while possibly inhibiting the secretion of the appetite-reducing hormone, melanocyte-stimulating hormone (α-MSH). Muscle Tissues: Studies on murine models with thermal injuries suggested that GHRP-2 may reduce pro-inflammatory markers and the activity of ubiquitin ligases, which are associated with muscle wasting.
